Monopolar spindle 1 kinase (MPS1), also known as TTK protein kinase, is a critical enzyme involved in cell division. Primarily, MPS1 plays a pivotal role in the spindle assembly checkpoint (SAC), a safety mechanism ensuring that chromosomes are properly aligned and attached to the spindle apparatus before cell division proceeds. This ensures accurate chromosome segregation and prevents aneuploidy, a condition associated with various diseases, including cancer. MPS1’s function in checkpoint signaling makes it an attractive target for cancer therapy. Inhibitors targeting MPS1 are being explored to potentially disrupt cancer cells' ability to correctly manage chromosome segregation, leading to cell death and tumor reduction.
