PGE synthase
Prostaglandin E synthase (PGE synthase) is a key enzyme involved in the biosynthesis of prostaglandin E2 (PGE2) from prostaglandin H2 (PGH2). PGE2 is a potent inflammatory mediator with diverse roles in pain perception, fever regulation, and inflammation. It also plays significant roles in modulating vascular tone and protecting the gastric mucosa. PGE synthase exists in multiple isoforms, including microsomal PGE synthase-1 (mPGES-1), which is often induced during inflammation. Due to its crucial role in generating PGE2 during inflammatory states, targeting PGE synthase, particularly mPGES-1, has become a promising strategy for developing anti-inflammatory drugs that can potentially offer relief in conditions like arthritis, pain, and fever without the side effects associated with traditional NSAIDs.
