ζ-Stat (Cat No.:I014807), also known as NSC37044, is a specific and atypical inhibitor of PKC-ζ (protein kinase C-zeta). It has an IC50 of 5 μM against PKC-ζ. ζ-Stat has demonstrated the ability to reduce proliferation and induce apoptosis in melanoma cell lines, showcasing potential antitumor activity in vitro. It shows limited inhibition on PKC-ι but exhibits significant inhibition on PKC-ζ. Treatment with ζ-Stat results in decreased cell proliferation in SK-MEL-2 and MeWo cells with increasing concentrations. Combining ζ-Stat with 5-FU has shown a synergistic effect in reducing the viability of LoVo CRC cells. ζ-Stat treatment also leads to a reduction in phosphorylated and total PKC-ζ, Bcl-2, and PARP levels, while increasing Caspase-3 and cleaved-PARP levels in melanoma cells
