Afatinib (BIBW2992) Dimaleate is an irreversible tyrosine kinase inhibitor targeting the ErbB family, including EGFR (HER1) and HER2. It binds covalently to the ATP-binding site, blocking phosphorylation and downstream signaling pathways that drive cell proliferation and survival. Afatinib is approved for the treatment of non-small cell lung cancer (NSCLC) with EGFR mutations and is being explored in other cancers involving ErbB overexpression. Its ability to inhibit multiple ErbB receptors makes it a promising option for targeted cancer therapy.
