CU-CPT-8m (CAT: I019896) is a specific TLR8 antagonist with an IC50 of 67±10 nM. It shows negligible cytotoxicity and directly recognizes TLR8 in cells. Notably, it does not affect TLR7 signaling at concentrations up to 75 μM. CU-CPT-8m inhibits the proinflammatory response in TLR8-overexpressing cells and abolishes the elevation of TNF-α and IL-8 mRNA levels induced by R848. In differentiated THP-1 monocytes, it dose-dependently inhibits R848-induced TNF-α production with an IC50 of 90±10 nM. CU-CPT-8m demonstrates its potential as a selective TLR8 antagonist for modulating immune responses.
