CU-CPT-9a(CAT: I019876) is a specific antagonist of Toll-like receptor 8 (TLR8). It exhibits a high potency with an IC50 value of 0.5±0.1 nM. CU-CPT-9a effectively reverses the elevation of downstream protein levels induced by R848, a TLR8 agonist, in a dose-dependent manner. Notably, CU-CPT-9a does not significantly affect the expression levels of TRIF and IRF3, which are associated with TLR4 and TLR3 signaling pathways, suggesting its specific targeting of TLR8. CU-CPT-9a has shown promise in suppressing TNF-α levels, aligning with the involvement of TLR8 in autoimmune diseases.
