GTP-14564(CAT: I007132) is a cell-permeable, reversible, and ATP-competitive tricyclic benzofurano-indazolo compound that acts as a potent and specific inhibitor of class III receptor tyrosine kinases, including c-Fms, c-Kit, and Fms-like tyrosine kinase 3 (FLT3). It exhibits inhibitory activity against both wild-type FLT3 (wt-FLT3) and FLT3 with internal tandem duplication (ITD-FLT3) mutations, with an IC₅₀ of approximately 0.3 µM. GTP-14564 has been shown to inhibit the growth of interleukin-3-independent Ba/F3 cells expressing ITD-FLT3 at 1 µM, while a 30-fold higher concentration is required to inhibit FLT3 ligand-dependent growth of Ba/F3 cells expressing wt-FLT3. Given its mechanism of action, GTP-14564 is most relevant to Cancer Disease Research, particularly in the study of acute myeloid leukemia (AML) where FLT3 mutations are implicated in disease progression.
