MOCAc-PLGL(Dpa)AR(Cat No.:M096430), also known as Mca-Pro-Leu-Gly-Leu-Dpa-Ala-Arg-NH₂, is a positively charged fluorogenic substrate utilized to assess the activity of matrix metalloproteinases (MMPs), specifically MMP-2, MMP-7, and MMP-9. The peptide sequence incorporates a 7-methoxycoumarin-4-yl (Mca) group at the N-terminus, serving as a fluorescent donor, and a 2,4-dinitrophenyl (Dnp) group attached to a diaminopropionic acid (Dpa) residue, acting as a quencher. Upon cleavage between the glycine and leucine residues by active MMPs, the fluorescence is restored, allowing real-time monitoring of enzyme activity. This substrate exhibits excitation and emission maxima at 328 nm and 393 nm, respectively. It is widely employed in biochemical assays to investigate MMP functions and screen potential MMP inhibitors.
