MRT 10(CAT: I010986) is a potent Smoothened (Smo) receptor antagonist with an IC 50 of 0.5 μM in HEK293 cells transiently expressing mouse Smo. It effectively inhibits Sonic Hedgehog (ShhN)-induced Gli luciferase reporter activity in Shh-light 2 cells with an IC50 of 2.5 μM. Additionally, it functions as an inverse agonist during nontranscriptional activation of the Hedgehog signaling pathway. MRT 10 is a valuable tool for studying Hedgehog pathway regulation and its implications in cancer and developmental disorders, offering insights into novel therapeutic strategies targeting Smo-dependent mechanisms.
