​NITD-688（Cat No.:I033344）is an orally active, pan-serotype inhibitor targeting the nonstructural protein 4B (NS4B) of the dengue virus (DENV). It exhibits potent antiviral activity against all four DENV serotypes, with EC₅₀ values ranging from 8 to 38 nM. In preclinical studies, NITD-688 demonstrated excellent efficacy; for instance, infected AG129 mice treated orally at 30 mg/kg twice daily for 3 days showed a 1.44-log reduction in viremia. Pharmacokinetic evaluations in rats and dogs revealed a long elimination half-life and good oral bioavailability. Mechanistically, NITD-688 binds directly to NS4B, disrupting its interaction with NS3, thereby inhibiting viral replication. These findings position NITD-688 as a promising preclinical candidate for dengue treatment.
