NSC-687852 (Cat No.:I000111), also known as b-AP15, is a specific inhibitor of the deubiquitinating enzymes UCHL5 and Usp14 within the 26S proteasome. It effectively blocks the deubiquitinating activity of the 26S proteasome and inhibits the function of two regulatory-particle-associated deubiquitinases, UCHL5 and USP14, resulting in the accumulation of polyubiquitin. b-AP15 induces tumor cell apoptosis independent of TP53 status and overexpression of the apoptosis inhibitor BCL2. In multiple myeloma cell lines and solid tumor models, b-AP15 demonstrates caspase-dependent and caspase-independent apoptotic effects. Additionally, b-AP15 has shown inhibitory activity against the selenoprotein thioredoxin reductase (TrxR). In various in vivo models, b-AP15 exhibits tumor growth inhibition, organ infiltration suppression, and prolonged survival. Combination therapies involving b-AP15 have demonstrated synergistic anti-multiple myeloma activity.
