QM31(CAT: I017407), also known as SVT016426, is a cytoprotective agent that acts as a selective inhibitor of Apaf-1. It effectively inhibits the formation of the apoptosome, which is the caspase activation complex composed of Apaf-1, cytochrome c, dATP, and caspase-9. With an IC50 of 7.9 μM, QM31 hinders the assembly of the apoptosome, thereby preventing caspase activation. Additionally, QM31 exhibits mitochondria protective properties and interferes with the intra-S-phase DNA damage checkpoint. In HeLa S100 cells, SVT016426 inhibits Apaf1-induced activation of caspase activity with an IC50 of 5 μM. These findings suggest the potential of QM31 as a therapeutic agent for modulating apoptotic pathways and protecting against cellular damage.
