RB-3(CAT: I035455) is a novel and selective polycomb repressive complex 1 (PRC1) inhibitor that directly targets the RING1B–BMI1 complex. By disrupting its interaction with nucleosomes, RB-3 effectively reduces global histone H2A ubiquitination (H2AK119ub), a key epigenetic modification involved in gene silencing. This disruption leads to the reactivation of differentiation pathways and suppresses self-renewal in acute myeloid leukemia (AML) cells. RB-3 shows significant potential in oncology research, particularly in studying epigenetic regulation of cancer and developing targeted therapies for leukemia by promoting the differentiation and apoptosis of malignant cells.
