RU.521 (CAT: I018751) is a highly potent and selective inhibitor of cyclic GMP-AMP synthase (cGAS), a key enzyme in the cellular innate immune response. By specifically targeting cGAS, RU.521 effectively hinders the activation of cGAS-mediated interferon signaling pathways. It significantly reduces the upregulation of interferon production in response to double-stranded DNA (dsDNA) activation. With an IC50 of 700 nM, RU.521 effectively suppresses dsDNA-induced reporter activity. Moreover, RU.521 showcases its therapeutic potential by attenuating the constitutive interferon expression in macrophages derived from an Aicardi-Goutières syndrome (AGS) mouse model.
