TCO-NHS ester(Cat No.:I016623) is an alkyl/ether-based linker commonly employed in the synthesis of proteolysis-targeting chimera (PROTAC) molecules. TCO-NHS ester contains a terminal trans-cyclooctene (TCO) group and an N-hydroxysuccinimide (NHS) ester functionality. The NHS ester enables efficient coupling with primary amines, such as those found in target protein binders, while the TCO group allows for bioorthogonal click chemistry reactions with tetrazine-functionalized molecules. This linker enables the conjugation of target protein binders to E3 ligase ligands, facilitating the selective degradation of specific proteins through PROTAC-mediated mechanisms. TCO-NHS ester contributes to the design and development of novel PROTAC molecules for targeted protein degradation applications.
