TRV-120027 (also known as oliceridine) is a novel G-protein biased ligand at the μ-opioid receptor, developed as an analgesic for the treatment of acute pain. Unlike traditional opioids, which activate both G-protein signaling and β-arrestin pathways, TRV-120027 selectively engages the G-protein pathway, which is associated with pain relief, while minimizing activation of the β-arrestin pathway, which is linked to adverse effects like respiratory depression and constipation. This selective activation aims to provide effective pain relief with a reduced risk of typical opioid side effects.
