UNC8732 is a second-generation NSD2 degrader (DC50 value of 60 nM; Dmax of 97%) that demonstrates improved NSD2 degradation and H3K36me2 reduction in cells. UNC8732 contains a primary alkyl amine, which is metabolized in cells to an active aldehyde species that promotes NSD2 degradation through the recruitment of FBXO22, a substrate recognition subunit of a Cullin 1 E3 ligase complex.
