ZT-12-037-01(CAT: I018735) is an inhibitor that targets STK19 and inhibits the oncogenic NRAS-driven melanocyte malignant transformation. It is an ATP-competitive inhibitor that can inhibit the phosphorylation of NRAS with an IC50 of 24 nM. ZT-12-037-01 significantly inhibits mutant NRAS-STK19-driven melanocyte colony formation and proliferation. It also has an inhibitory effect on STK19WT and STK19D89N-activated NRAS phosphorylation. In vivo, ZT-12-037-01 inhibits the growth of SK-MEL-2 xenograft melanoma and induces apoptosis by increasing cleaved caspase-3.
