This receptor binds the C-X-C chemokine CXCL12/SDF-1, triggering intracellular calcium ion increases and MAPK1/MAPK3 activation while engaging the AKT signaling cascade. It regulates cell migration, including during wound healing, and acts as a receptor for extracellular ubiquitin, enhancing calcium ion influx and reducing cellular cAMP levels. The receptor binds bacterial lipopolysaccharide (LPS), mediating LPS-induced inflammatory responses such as TNF secretion by monocytes.
It plays key roles in hematopoiesis, cardiac ventricular septum formation, and gastrointestinal vascularization through endothelial cell remodeling. Additionally, it contributes to cerebellar development and hippocampal neuron survival in the CNS (by similarity). In microbial infections, it functions as a coreceptor for HIV-1 X4 isolates (with CD4) and as a primary receptor for certain HIV-2 isolates, facilitating Env-mediated viral fusion.
