This protein is essential for the post-translational delivery of tail-anchored (TA) proteins to the endoplasmic reticulum (ER). In conjunction with GET1/WRB, it serves as a membrane receptor for soluble GET3/TRC40, which specifically binds the transmembrane domains of TA proteins in the cytosol, facilitating their ER targeting. It is also required for the stability of GET1.
Beyond its role in protein trafficking, it stimulates calcium signaling in T cells by aiding intracellular calcium elevation and is critical for the survival of peripheral follicular B cells (by similarity). These functions underscore its importance in maintaining cellular processes, immune signaling, and lymphocyte survival.
