This enzyme methylates CpG residues, preferentially targeting hemimethylated DNA, and plays a vital role in maintaining DNA methylation patterns essential for epigenetic inheritance. During the S phase, it associates with DNA replication sites to preserve methylation on newly synthesized strands, while in G2 and M phases, it maintains methylation independently of replication. This enzyme ensures the inheritance of methylation patterns established during development and coordinates DNA methylation with histone methylation.
It represses transcription by directly binding to HDAC2 and, in association with DNMT3B and CTCFL/BORIS, activates BAG1 expression by modulating promoter histone H3 dimethylation at H3K4 and H3K9. It forms corepressor complexes involved in KRAS-mediated promoter hypermethylation and silencing of tumor suppressor genes in colorectal cancer. Additionally, it maintains transcriptional repression in undifferentiated embryonic stem cells, silences tumor suppressor gene promoters, and promotes tumor growth, highlighting its role in development and cancer progression.
