The receptor tyrosine kinase EGFR binds ligands from the EGF family to activate signaling cascades that translate extracellular signals into cellular responses. Known ligands include EGF, TGFA/TGF-alpha, AREG, epigen (EPGN), betacellulin (BTC), epiregulin (EREG), and heparin-binding EGF (HBEGF). Ligand binding induces receptor homo- or heterodimerization and autophosphorylation on key cytoplasmic residues, enabling the recruitment of adapter proteins like GRB2, which initiate complex downstream signaling pathways.
EGFR activates at least four major signaling cascades: RAS-RAF-MEK-ERK, PI3K-AKT, PLCgamma-PKC, and STATs. It may also engage the NF-kappa-B pathway. Additionally, EGFR directly phosphorylates proteins such as RGS16, enhancing its GTPase activity to potentially link EGFR and G protein-coupled receptor signaling. It also phosphorylates MUC1, facilitating its interaction with SRC and beta-catenin (CTNNB1).
EGFR promotes cell migration through interaction with CCDC88A/GIV, retaining the receptor at the cell membrane post-ligand stimulation and sustaining signaling for migratory responses. Beyond cellular signaling, EGFR contributes to learning and memory performance and mammalian pain signaling, particularly in long-lasting hypersensitivity. These diverse roles underline EGFR’s critical functions in cellular communication, migration, and broader physiological processes.
