The protein tyrosine kinase ERBB2 is a component of several cell surface receptor complexes but requires a coreceptor for ligand binding. It is an essential part of the neuregulin-receptor complex, though neuregulins do not bind ERBB2 directly. GP30 is a potential ligand for this receptor. ERBB2 regulates peripheral microtubule outgrowth and stabilization. Upon activation, the MEMO1-RHOA-DIAPH1 signaling pathway inhibits GSK3B through phosphorylation at the cell membrane, preventing the phosphorylation of APC and CLASP2. This enables their membrane association, facilitating MACF1 localization to the membrane, which is critical for microtubule capture and stabilization.
In the nucleus, ERBB2 contributes to transcriptional regulation. It associates with the 5′-TCAAATTC-3′ sequence in the PTGS2/COX-2 promoter to activate transcription. It also participates in the transcriptional activation of CDKN1A, a process involving STAT3 and SRC. Additionally, ERBB2 supports rRNA gene transcription by RNA Pol I, enhancing protein synthesis and promoting cell growth. Its roles in signaling and transcription underscore its significance in cellular regulation.
