MET is a receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding hepatocyte growth factor (HGF). It regulates key physiological processes, including cell proliferation, scattering, morphogenesis, and survival. Upon ligand binding, MET undergoes autophosphorylation on its intracellular domain, creating docking sites for downstream signaling molecules such as PIK3R1, PLCG1, SRC, GRB2, STAT3, and GAB1.
These interactions activate multiple signaling pathways, including RAS-ERK, PI3K-AKT, and PLCgamma-PKC. The RAS-ERK pathway is linked to morphogenesis, while PI3K-AKT supports prosurvival effects. During embryonic development, MET signaling plays vital roles in gastrulation, neuronal precursor migration, angiogenesis, and kidney formation. In skeletal muscle development, it facilitates the migration of muscle progenitor cells and the proliferation of secondary myoblasts.
In adults, MET contributes to wound healing, organ regeneration, and tissue remodeling. It also supports the differentiation and proliferation of hematopoietic cells and may regulate cortical bone osteogenesis. MET’s diverse roles highlight its importance in development, tissue repair, and maintaining physiological homeostasis.
