Hypoxia-inducible factor (HIF) is a transcription factor that regulates oxygen-responsive genes. It forms a heterodimer with ARNT, binding to the core DNA sequence 5′-TACGTG-3′ in hypoxia response elements (HRE) of target gene promoters. HIF regulates vascular endothelial growth factor (VEGF) expression, playing a crucial role in blood vessel development, lung tubular system formation, and potentially the establishment of the blood-brain barrier.
HIF strongly activates Tie-2 tyrosine kinase expression, requiring transcriptional coactivators such as CREBBP and possibly EP300. Interaction with the redox regulatory protein APEX1 further activates its C-terminal transcriptional activation domain (CTAD), enhancing its role in hypoxia signaling and oxygen homeostasis in various physiological processes.
