This low-affinity receptor binds neurotrophins such as NGF, BDNF, NTF3, and NTF4. When forming a heterodimer with SORCS2, it binds precursor forms of NGF, BDNF, and NTF3 with high affinity, exhibiting lower affinity for their mature forms. The receptor plays a critical role in the differentiation and survival of specific neuronal populations during development and can mediate both neural cell survival and cell death. It also contributes to the inactivation of RHOA.
In metabolic regulation, it influences insulin-dependent glucose uptake by modulating RAB31 activity, aiding GLUT4 translocation to the cell surface in adipocytes and skeletal muscle cells. Additionally, it is essential for circadian oscillation of clock genes in the brain’s suprachiasmatic nucleus and liver, as well as genes regulating glucose and lipid metabolism in the liver.
