Tyrosine-protein kinase FLT3 functions as a cell-surface receptor for the cytokine FLT3 ligand (FLT3LG), playing a pivotal role in the differentiation, proliferation, and survival of hematopoietic progenitor and dendritic cells. Upon activation, FLT3 promotes phosphorylation of SHC1 and AKT1, triggering downstream effectors such as MTOR. It also activates RAS signaling, leading to the phosphorylation of MAPK1/ERK2 and MAPK3/ERK1, and promotes phosphorylation of proteins including FES, FER, PTPN6/SHP-1, PTPN11/SHP-2, PLCG1, and STAT5A/B. Wild-type FLT3 activation marginally stimulates STAT5 signaling, whereas mutations inducing constitutive kinase activity drive uncontrolled cell proliferation and resistance to apoptosis by activating multiple pathways. These properties underline FLT3’s importance in normal hematopoiesis and its role in the pathogenesis of hematological malignancies, making it a crucial therapeutic target.
