This tyrosine-protein kinase functions as a cell-surface receptor for VEGFA, VEGFB, and PGF, playing a crucial role in embryonic vascular development, angiogenesis, cell survival, migration, macrophage function, chemotaxis, and cancer cell invasion. It positively regulates postnatal retinal hyaloid vessel regression and may act as a negative regulator of embryonic angiogenesis by controlling excessive endothelial cell proliferation.
In adulthood, it supports endothelial cell proliferation, survival, and angiogenesis in a cell-type-specific manner. It promotes PGF-induced endothelial cell proliferation and cancer cell growth but does not stimulate normal fibroblast proliferation (in vitro). With high VEGFA affinity and relatively low kinase activity, it may regulate VEGFA signaling by sequestering VEGFA and forming heterodimers with KDR to modulate KDR signaling.
Ligand binding activates multiple pathways, including PLCG, which generates diacylglycerol and inositol 1,4,5-trisphosphate, activating protein kinase C. It phosphorylates PIK3R1, triggering phosphatidylinositol kinase activity and downstream signaling. Additionally, it activates the MAPK/ERK and AKT1 pathways, phosphorylating AKT1 at ‘Ser-473’. It also mediates phosphorylation of SRC, YES1, CBL, and PTK2/FAK1. These diverse roles highlight its importance in vascular biology, cancer progression, and cellular signaling regulation.
